Background: Aortic banding was performed as previously described (Rockman et al, PNAS 88:8277, 1991). Briefly, after general anesthesia and intubation, a small left-sternal thoracotomy was performed and aortic constriction accomplished by ligation of the aorta next to a 27 gauge needle, leaving a 0.4 mm lumen when the needle was removed. This approach produces a rapid increase in LV mass of approximately 35% after one week. Trans-ligation gradients were measured prior to sacrifice and hearts from animals with gradients of 60-70 mm Hg were used in these experiments. The heart was rapidly excised, the left ventricle was isolated by dissection and frozen in liquid nitrogen. Hearts were obtained after 1, 4 and 8 weeks of banding.
